A téma rövid leírása, a kidolgozandó feladat részletezése: 
A modern orvosi és műszaki tudományok együttműködésében kialakult határtudomány, a biomechanika feladata a szövetek, mint biológiai anyagok mechanikai tulajdonságának vizsgálata. Egyre fontosabbá válik a humán lágyszövetek anyagmodelljeinek lehető legpontosabb meghatározása és kísérletekkel történő verifikálása. A húzókísérletek alkalmasak a szövetminták különböző anyagmodellekkel leírható makroszkópikus anyagi viselkedésének leírására. Önálló részterület ebben a témában a kollagénnel, mint fő teherviselő struktúrával rendelkező szövetek mechanikai tulajdonságainak meghatározása, melyek különleges felépítésük révén egyszerre biztosítanak rugalmasságot és szilárdságot pl. az ínaknak, az ereknek, vagy akár a szaruhártyának. A BME és a SE évtizedek óta szoros együttműködésben végez nyúlásméréses vizsgálatokat és készít numerikus szimulációkat. A korábbi kutatások eredményeinek statisztikai elemzése során megfigyelhető szignifikáns eltérés, vagy éppen ezek hiánya veti fel a kutatás folytatásának igényét. A korábbiakban elvégzett, kizárólag mechanikai kísérletek mellett klinikai vizsgálatokkal alátámasztott méréssorozat elvégzése is szükséges. 
Transzplantáció és más műtéti eljárások elterjedésének következtében lényeges kérdés, hogy a sterilizálás, tárolás, fagyasztás és felolvasztás módja, illetve egyéb tulajdonságok (pl. nem, kor, hormonszintek), milyen mértékben változtatják meg a mechanikai tulajdonságokat. Ezek alapján pedig a hazánkban jelenleg alkalmazott műtéti protokollok optimalizálása is lehetségessé válik, illetve a nemzetközi versenyképesség is javul. Nem csupán az egyes alcsoportok jellegzetes eltérések megállapítása a célunk, hanem az okok minél pontosabb felderítése. Mindehhez szükséges a humán minták patológiai elemzése, a minták további tárolása pedig egy olyan genetikai vizsgálatokra is alkalmas biobank részét tudja képezni, amely akár évtizedekkel később a humán DNS-sel kapcsolatos értékes információkkal szolgálhat, így össztársadalmi képet is nyújthat. 
A verifikált anyagmodellek a numerikus modellezések bemenő adataiként is szolgálnak. Az anyagi viselkedés megismerése révén az adott szerv működésének virtuális ikerrel történő nyomon követese is lehetséges. A különböző műtéti beavatkozások, mint például akár humán ér- vagy ín homograft transzplantációs műtétek numerikusan szimulálhatók, vagy akár emberi szaruhártya cérnával való rögzítése esetén a dioptriaváltozásra gyakorolt hatása jósolhatóvá, vagy egy aneurizma (zsákszerű értágulat) fejlődése is nyomon követhetővé válna numerikus úton is. A szövettani vizsgálatok során feltárt, a felépítésben dokumentált jellegzetességek (pl. száleloszlás, szálirány és szálmennyiség) mezo-szintű numerikus szimulációt is lehetővé tesznek. Lokális elváltozások során indokolt lehet a makroszintű vizsgálatok ilyen módon történő bővítése. Mikroszerkezeti szinten pedig maguk a kollagénszálak is vizsgálhatók. A kollagénszálak elrendezése és felépítése is klasszikus mérnöki kérdéskört idéz, felépítésük ugyanis kísértetiesen emlékeztet a mérnöki szerkezeteknél gyakran használt sodronykötelekre, többszintű hierarchikus felépítést mutatva. A legalsóbb szintet egy tripla hélix jelenti, melyet molekuláris dinamikai szimulációkkal lehetséges modellezni. A mikroszerkezeti szintről való építkezés során homogenizációs eljárásokkal a makroszerkezeti szintig el lehet jutni, mely jellemzők jelen PhD témakiírás esetében meghatározásra kerülnek, így a cél is adottá válik. 
Az elvégzendő feladatok: 
    • Kollagén szövetek erőelmozdulás diagramjainak meghatározása, a mechanikai modellhez szükséges anyagi paraméterek számítása, a páciensek anamnézise alapján statisztikai módszerekkel, a különböző alcsoportok összehasonlítása. 
    • A makroszerkezeti vizsgálatok során kapott eredmények mikroszerkezeti, szövettani megfigyelésekkel történő verifikálása. 
    • Numerikus modellek készítése a kollagén szövetek makro-, mezo- és mikroszerkezeti szintjén.

The task of biomechanics, a frontier science between modern medical and technical sciences, is to examine the mechanical properties of tissues as biological materials. It is becoming more and more important to determine the material models of human soft tissues as precisely as possible and to verify them with experiments.   Tensile tests are suitable for describing the macroscopic material behavior of tissue samples, described with different material models. An independent sub-field in this topic is the determination of the mechanical properties of tissues with collagen as the main load-bearing structure (e.g. tendons, blood vessels, or even the cornea), which, due to their special structure, provide flexibility and strength at the same time. For decades, BME and SE have been conducting strain measurement tests and numerical simulations in close cooperation. Significant deviations observed during the statistical analysis of the results of previous researches, or the absence of them, raises the need to continue the research. In addition to the previously performed, exclusively mechanical experiments, it is also necessary to carry out a series of measurements supported by clinical tests.  
 
As a result of the spread of transplantation and other surgical procedures, an important question is to what extent sterilization, storage, freezing and thawing methods, as well as other properties (e.g. gender, age, hormone levels), change the mechanical properties. Thus it becomes possible to optimize the surgical protocols currently used in our country, and the international competitiveness also improves. Our aim is not only to establish the characteristic differences of each subgroup, but to find out the causes as precisely as possible. For all of this, the pathological analysis of human samples is necessary, and the further storage of the samples can form part of a biobank suitable for genetic tests, which can provide valuable information about human DNA even decades later, thus an overall picture of society.  
 
The verified material models also serve as input data for numerical modeling. By getting to know the material behavior, it is also possible to monitor the functioning of the given organ with a virtual twin. Various surgical interventions, such as human vessel or tendon homograft transplantation operations, can be numerically simulated, or even in the case of fixation of the human cornea with a thread, its effect on the diopter change can be predicted, or the development of an aneurysm can also be tracked numerically. The characteristics revealed during the histological examinations and documented in the structure (e.g. fiber distribution, direction and quantity) also enable meso-level numerical simulation. In case of local changes, it may be justified to expand the macro-level examinations in this way. At the microstructural level, the collagen fibers themselves can also be examined. The arrangement and structure of the collagen fibers also evoke a classic engineering question, as their structure resembles of the wire ropes often used in engineering, showing a multi-level hierarchical structure. The lowest level is represented by a triple helix, which can be modeled by molecular dynamics simulations. During construction from the microstructural level, homogenization procedures can be used to reach the macrostructural level, which characteristics are determined in the case of this PhD topic announcement, so the goal becomes known.
 
In the research program the following tasks should be performed:
    • Determine force-displacement diagrams of collagen tissues, calculate the material parameters for the mechanical models, and compare the different subgroups using statistical methods based on the patients' anamnesis. 
    • Verify the results obtained during the macrostructural tests with microstructural and histological observations. 
    • Create numerical model(s) of the collagen tissues on the macro-, meso- and micro-structural level.
